My laboratory has been engaged in the Pathogenesis and Etiology of Nasopharyngeal Carcinoma (NPC). In endemic regions, NPC presents as a complex disease caused by an interaction of Epstein-Barr virus (EBV) chronic infection, environmental, and genetic factors, in a multistep carcinogenic process. Currently, we are interested in the mechanism of EBV infection and transformation of nasopharyngeal epithelial cells. We have previously identified the genetically determined susceptibility of NPC patients and the variation of EBV, furthermore, completed the first full-length sequence analysis of an NPC-derived EBV strain. We have already established the first cellular proto-oncogene immortalized nasopharyngeal epithelial cell line, and we demonstrated the functional and mechanistic links between the oncoprotein Bmi-1 and the tumor suppressor PTEN in the development and progression of cancer. Most importantly, we recently established anin vitroEBV infection epithelial model with highly infective efficiency. Moreover, we discovered that LMP2A induces EMT and stem-like cell self-renewal in NPC, suggesting a novel mechanism by which Epstein-Barr virus induces the initiation, metastasis and recurrence of NPC.
